Aim: The aim of the present dissertation is to identify the role of POPDC proteins in
progression of various cancers.
Introduction (function, structure, role) (Amunjela, 2017)
POPDC proteins are members of POPDC gene family. There are 3 members namely
POPDC1, POPDC2 and POPDC3 encoding for POPDC1 or BVES (Blood Vessel
Epicardial), POPDC2 and POPDC3 proteins respectively. These proteins are
transmembranous in nature. These dimer proteins get inserted into the cell membrane and are
held by a disulphide bond. POPDC1 has been identified as involved in malignant processes
and represents a less ubiquitous binding target for cAMP (Amunjela, 2017).
Structure
Transmembranous protiens encoded by POPDC genes are medium sized with short
amino end terminal comprising of 27-39 residual units. Gene mapping reveals one or two
functional N-glycosylation sites. POPDC undergoes extensive glyosylation. Major form of
POPDC protein is POPDC1 with an apparent weight of 55-58kDal. The 151 amino acid long,
Popeye domain is unique as it is found only in POPDC1. Proteins that are non PODC in
nature have CAPs or catabolite activator proteins as major form with maximum similarity
sequence. CAPs are also known as cAMP receptor proteins. Structurally cAMP and popeye
domain have evidently shown major similarities. A family of cAMP binding proteins known
as phospodiesterases deactivate cAMP which are involved in conversion of cAMP to AMP.
In mammalian cells, five known effector molecules bind directly to cAMP. They are EPAC,
